Clearance is volume of plasma cleared of a drug per unit time; more the clearance less the half life. Hepatic metabolism will affect hepatic clearance. Hepatic clearance
= ER . HBF. Total clearance= CLr + CL h+ CLx. Volume of distribution will affect the clearance inversely. Eg. Fentanyl Vd more than alfentanyl, so cleared less.
